The seat of Lycoming County, Williamsport lies 131 miles northwest of Philadelphia, 166 miles east of Pittsburgh, and 85 miles almost due west of Scranton. A major crossroads, the city is located on U.S. Route 220, a major North-South highway established in 1926 that runs through the middle of Pennsylvania and continues down to North Carolina. Once the highway hits Williamsport, it overlaps with U.S. 15 downtown before splitting near Halls Station. Historically, its location along the West Branch of the Susquehanna River first put Williamsport on the map as a logging community. Great logs were tied together into rafts and floated South. Over time, the city found itself on the West Branch of the Pennsylvania Canal system, and became a transfer station for the Reading Railroad, the New York Central, and the Pennsylvania Railroad. Although passenger service is no longer offered, freight continues to move onboard the Lycoming Valley Railroad, centered in Williamsport, connecting to Norfolk Southern and Canadian Pacific lines.

The Peter Herdic Transportation Museum delves into the history of transportation in the area, and how it impacted the lives of Lycoming County residents. Exhibits include a prehistoric, Native American birch-bark canoe, a fully restored 1962 GMC bus, and one of only two remaining 1949 Pennsylvania Pullman Parlor railroad cars.

The West Branch of the Susquehanna River played a significant role in the history of the city, and visitors can learn more during a dinner cruise on the Hiawatha Paddlewheel Riverboat. A guide will point out spots of interest, comment on the history of the area, explain Colonial-era and early-American lumbering, and outline the role the Susquehanna River played in the region and in the big push to settle the West. The Susquehanna River Walk is a paved trail that winds along the banks of the river and offers opportunities to walk, bike, birdwatch, and fish. Perhaps as a remnant of its early history as a station along the Underground Railroad, Williamsport racially integrated its schools comparatively early, in 1948.

The city of Williamsport sprawls along the Northern edge of the Susquehanna, with many businesses, restaurants, shops, and health care organizations located within walking distance of its shoreline. The Williamsport Regional Medical Center is currently managed by the University of Pittsburgh Medical Center (UPMC) – Susquehanna, which began as an alliance between three hospitals: Williamsport, Muncy Valley Hospital, and Divine Providence. It includes the Soldiers & Sailors Memorial Hospital in Wellsboro, and manages additional medical facilities in Sunbury, Montoursville, and Lock Haven.
